Abstract: CD-TEC包接法應用於Bacillus macerans(IAM 1243)之環狀糊精葡萄糖?基移轉?(CGT)活性檢測之條件為:2%可溶性澱粉基質,pH 6.0,反應時間2小時;偶合反應活性單位與包接法活性單位比較,當前者為1.5活性單位以下時有直線關係。
探討數種單醣、雙醣或可溶性澱粉為碳源,均無包接法可偵測之最低活性倍數呈現,只有麥麩及米糠可呈現,又以麥麩效果較佳;加葡萄糖於培養基中對CGT之生產有抑制現象;此菌培養於無機或有機氮源中皆可產生CGT。
經實驗發現CGT之最佳生產培養機組成為每公升培養基中含果糖10g,麥麩40g,聚胜?5g,MgSO4 .7H2 O0.3g;在500mL三角瓶中培養之最適條件為:培養液20mL,培養溫度45℃,培養初pH值7.0,振盪速率200rpm,培養時間24小時。
